Humanitas Advanced Optical Microscopy Unit (HAOUM) (https://www.humanitas-research.org/facility/advanced-optical-microscopy/) provides access to state-of-the-art light microscopes for scientists and clinicians within and outside ICH. The facility is in charge for set up and maintenance of instrumentation and training, development of methods and protocols and in vivo experimental models for confocal and IVM analysis. The personnel of the facility are involved in scientific collaborations for the development of optical microscopy-based applications for biomedical research. HAOUM has an independent research program. HAOUM is based on multi-scale Imaging approach through the use of technologies, including confocal microscopy, STED-based super-resolution, FLIM/FCS, multiphoton microscopy, in vivo optical Imaging based on luminescence and fluorescence tomography, chemical clarification of organs and 3D analysis, and high-throughput Imaging on tissues. High-capacity network of digital workstations is available for data processing and image analysis.
The knowledge accumulated in the context of super-resolution optical microscopy represents the starting point and main prerequisite for the growth strategy and technological direction of the new Innovation Building in the Humanitas University campus. The new challenge will be to start from near-molecular super-resolution and move beyond the nanoscale toward the study of molecular localization and structure in the physiological context by acquiring the CLEM technology. The CLEM core will include a workflow consisting of LSM 900 with Airyscan 2, ZEISS; high-pressure freezing, CryoCapCell; CryoFIB-SEM (Crossbeam) XB 550, ZEISS; TEM, Jeol; GeminiSEM 360, ZEISS.
We are looking for a Senior Operator, Cryo-Electron Microscopy.
The person filling this position will:
• daily maintain daily instruments.
• provide design support for experiments.
• provide sample preparation and acquisition services.
• train and support user.
The suitable candidate is expected to have proven experience in use of EM including sample preparation, knowledge of microscope hardware, use of Imaging analytical software, basic knowledge of confocal microscopy. A minimum of 3 years of hands-on experience in EM is required as well as knowledge of ultra-structures found in cells, tissues, bacteria, and viruses.
